| In addition to the antitumor effect, PHY906 was also examined as a modulator of host cytotoxicity induced by CPT-11. Two factors were monitored in the experiments: body weight loss and mortality. |

| We observed a dose-dependent behavior of PHY906 on weight loss in CPT-11 treated mice (Figure 2). |

| CPT-11 treated animals receiving supplemental treatment with 500 mg/kg, bid, of PHY906 exhibited significant (p < 0.05) improvement in maintaining body weight and recovered their original body weight more rapidly. However, mice receiving 250 or 125 mg/kg, bid, of PHY906 showed no difference in body weight loss compared to controls. |

| Forty of 40 tumor-bearing mice (100%) survived treatment with PHY906 alone, indicating no or very low toxicity from PHY906. In contrast, treatment with CPT-11 alone resulted in only 36 of 45 tumor-bearing mice (80%) surviving after 8 days. However, this survival rate dramatically improved to 96% (24 of 25 mice) after receiving 4 days of PHY906 treatment in combination with CPT-11. This suggests that PHY906 treatment can protect mice against mortality induced by a single dose of 400 mg/kg CPT-11. |
